Indicated for the prevention and treatment of Vitamin D deficiency and as a supplement in patients with conditions requiring increased Vitamin D, such as osteoporosis, rickets, osteomalacia, and malabsorption syndromes. May be used as an adjunct in managing hypocalcemia.
Contraindicated in patients with hypercalcemia, hypervitaminosis D, malabsorption syndrome with abnormal sensitivity to Vitamin D toxic effects, or known hypersensitivity to any ingredient.
At recommended doses, adverse effects are rare. Excessive intake may lead to hypervitaminosis D, manifesting as hypercalcemia, hypercalciuria, nausea, weakness, and renal calcification. Allergic sensitization has been reported rarely.
Use with caution in patients with kidney disease or thyroid disorders. Monitor serum calcium levels with prolonged use. Do not exceed recommended dose. Keep out of reach of children. Store at 20°–25°C; protect from heat and light.
Cholecalciferol (Vitamin D3) undergoes two-step hydroxylation: first in the liver to 25-hydroxyvitamin D, then in the kidneys to the active 1,25-dihydroxyvitamin D (calcitriol). Active metabolites promote intestinal absorption of calcium and phosphorus, regulate bone mineralization, and modulate parathyroid hormone secretion.
Store at 20°–25°C (68°–77°F); excursions permitted to 15°–30°C (59°–86°F). Protect from heat and light. Keep out of reach of children.
